毛细管电泳法测定黄瓜和西红柿中的谷胱甘肽 被引量:6

摘要 在未涂层的石英毛细管中,以0 04mol/LNa2HPO4 0 04mol/LNaH2PO4的混合体系(pH7 32)为运行缓冲液,采用紫外检测波长为200nm,建立了果蔬中谷胱甘肽(GSH)的毛细管电泳测定方法。结果表明:GSH为3 07~307mg/L时具有良好的线性关系,相关系数r=0 9995;最低检测质量浓度为1 54mg/L(S/N=3)。利用该方法对黄瓜及西红柿中的GSH进行了测定,在3个添加水平上进行加标回收试验,结果GSH在西红柿中的加标回收率分别为99 04%,94 96%和101 4%,相对标准偏差(RSD)分别为1 53%,3 78%和1 89%;GSH在黄瓜中的加标回收率分别为91 25%,94 82%和99 35%,RSD分别为1 46%,3 47%和4 00%。该方法具有快速、简便、灵敏、重现性好等特点。 A method for the determination of glutathione (GSH) in tomatoes and cucumbers by capillary electrophoresis with UV detection (200 nm) was developed. After simple treatment of the samples, separation and determination of GSH was obtained within 7 min by using the phosphate buffer system containing 004 mol/L Na2HPO4004 mol/L NaH2PO4 (pH 732). The quantitative analysis was performed with external standard under the optimal conditions. The calibration curve of GSH showed good linearity in the range of 307-307 mg/L with r=0999?5, and the detection limit was 154 mg/L (S/N=3). For spiked tomato samples with standard at three levels (9219, 1229 and 1537 mg/L), the recoveries were 9904%, 9496% and 1014%, and the RSDs were 153%, 378% and 189%, respectively. For cucumbers, the recoveries were 9125%, 9482% and 9935%, and the RSDs were 146%, 347% and 400%, respectively. The method has the advantages of being simple, convenient and sensitive. The method can be used for the routine analysis of GSH in fruits and vegetables.
