摘要
通过80 %饱和度硫酸铵沉淀、DEAE -SephadexA -50离子交换柱层析和SephadexG -100凝胶过滤柱层析等方法从Aspergillussp .1 -13菌株的白酒丢糟固体培养物的抽提液中分离纯化到一种 β-木糖苷酶。通过SDS -PAGE分析测定其分子量为110.9KD。该酶最适反应pH值为3.2~3.8之间,最适反应温度为70℃。该酶具有较宽的 pH稳定性 ,并且在60℃以下较稳定。
By(NH4)2SO4fractionation and chromatography on DEAE-Sephadex A-50 and Sephadex G-100 colums, one β-D-xylosidase was purified from a crude culture fitrate of Aspergillus sp., grown on milled distiller's grain in solid state. The molecular weight of the β-D-xylosidase was 110.9KD as determined by SDS-PAGE. The optimum pH was 3.2~3.8. The optimum temperature was 70℃. It was stable in the pH range 3.0~9.0 and below 60℃.
