摘要
探讨采用当前国际上较先进的DNA损伤检测技术-单细胞凝胶电泳技术(SCGE,又名彗星试验),来检测农药对小鼠DNA的损伤,尝试建立小鼠的单细胞凝胶电泳技术来评价农药对生态环境和人体健康的影响.结果表明两种新型杀虫剂吡虫啉和抑食肼在一定的剂量范围内对小鼠的DNA均有一定程度的损伤作用,各浓度组与阴性对照组相比具有显著性差异(P<0.01),且具有明显的剂量效应关系.认为小鼠单细胞凝胶电泳技术能敏感地检测农药对细胞DNA的损伤,并认为细胞的DNA损伤可作为农药等化学品致突变性检测的生物标志物.
In this paper, we estimate the damage caused by two new pesticides, Imidacloprid and RH-5849, to DNA of mice peripheral blood lymphocytes and seminal cells by using alkaline single cell gel electrophoresis assay (SCGE, namely comet assay). The method of SCGE of mice peripheral blood lymphocytes or seminal cells was established to assess the risks of pesticides or other chemicals to the eco-environment and human health. The results showed that the damage of DNA caused by the two pesticides was dose dependent, and significant differences of DNA damage were shown between the treated animal groups and the negative control groups (P< 0.01). The SCGE was a very sensitive method in determining the damage of DNA, and the damage of DNA could be considered as a biomarker in determining the environmental mutagens.
