摘要 采用响应面方法对康氏木霉(Trichodermakoningii)ZJ5生产纤维素酶的培养基进行了优化.首先通过全因子实验分析培养基组分;稻草粉、麦麸、大麦粉和(NH4)2SO4对纤维素酶三个组分活性的影响,确定主要影响因子为稻草粉和(NH4)2SO4,前者为正影响,后者为负影响,用最陡爬坡路径逼近最大响应区域.利用中心组合设计及响应面分析确定主要影响因子的最佳浓度.在优化培养基中发酵6d,CMC酶活、滤纸酶活和β-葡萄糖苷酶活分别达到了765.8、155.3U/mL和39.54U/mL. Response surface methodology was used to optimize a medium for cellulase production of Trichoderma koningii ZJ5. In the first optimization step the effect of straw powder,barley flour,wheat bran and (NH4)2SO4 on cellulase production was evaluated using a full factorial design. Straw powder had positive effect,while (NH4)2SO4 had negative effect on cellulase production. The path of steepest ascent was used to approach the optimal region of the medium composition. The optimal concentrations of straw powder and (NH4)2SO4 were determined by a central composite design and response surface analysis.The maximum activity of carboxymethyl cellulose enzyme (CMCase),filter paper enzyme (FPA) and β- glucosidase is 765.8 U/mL,155.3 U/mL and 39.54 U/mL,respectively under the optimized culture medium after 6 days.
