摘要 目的:建立羟丙哌嗪对映体拆分的高效液相色谱法。方法:使用Chiracel-OC手性柱,以异丙醇-正己烷(15:85)为流动相,流速0.5ml/min,检测波长250 nm;将羟丙哌嗪酯化衍生后进样拆分。结果:在正相条件下,上述衍生物对映体实现基线分离,分离度为4.0,理论板数均大于6000,左、右旋异构体衍生物峰面积比值精密度(RSD)为0.63%。结论:方法简便,精密度高,可用于羟丙哌嗪的手性拆分。 Objective: To develop a chiral HPLC method for the determination of the two enantiomers of dropropizine, a novel antitussive drug. Methods: Using a commercial available chiral stationary phase (CSP) Chiracel-OC column, with the mobile phase of isopropanol-hexane (15: 85 ) , at the flow rate of 0. 5 ml/min and the detection wavelength of 250 nm, the racemic drug esterified with acetic an-hydride was separated. Results: The resolution was found to be 4. 0 with high column efficiency (over 6 000 theoretical plates) to achieve base-line separation. The RSD of the ratio of the areas of two enantiomers was 0. 63%. Conclusion: The present study provides a simple and reproducible method for the enantiomeric separation of dropropizine.
