葛根素注射液对实验大鼠糖尿病牙周炎的干预作用及血清AGEs、IL-6、TNF-α表达的影响

Intervention Effects of Puerarin Injection (葛根素注射液) on Rats with Diabetic Periodontitis and Influences on Serum AGEs ,IL-6 and TNF-α Expressions

目的:观察葛根素对实验大鼠糖尿病牙周炎的干预效果及血清AGEs(晚期糖基化终末产物)、IL-6(白细胞介素6)及TNF-α(肿瘤坏死因子α)表达的影响。方法:100只SD大鼠,随机选取20只为正常组,其余80只进行糖尿病牙周炎建模,将成模的60只大鼠随机分为模型组、阳性(二甲双胍)对照组、葛根素组,每组20只。分组给药6周,每组取10只大鼠,观察牙周组织病理变化及牙槽骨丧失程度;另外10只取血清ELISA法检测AGEs、IL-6、TNF-α表达。结果:与模型组比较,葛根素组大鼠牙周组织病理变化及牙槽骨丧失明显减轻,作用于二甲双胍阳性组相当。与正常组比较,模型组大鼠血清AGEs、IL-6、TNF-α水平明显升高(P<0.05),葛根素组及二甲双胍组给药6周后三者水平明显降低,与模型组比较具有统计学意义(P<0.05);两给药组作用相近,差异无统计学意义。结论:葛根素对大鼠糖尿病牙周炎有一定干预效果,其作用机制与减少血清AGEs积聚,降低IL-6、TNF-α等炎性介质表达有关。

Objective: To observe the intervention effects of Puerarin Injection on rats with diabetic periodontitis and influences on their serum AGEs,IL-6 and TNF-α expressions. Methods: One hundred Sprague-Dawley rats were selected and 20 of them were randomly selected as normal control group,the other 80 rats were modeled for diabetic periodontitis, 60 rats with successful modeling were randomly divided into model, positive control(metformin) and Puerarin group, 20 rats in each group,they were given saline,metformin and Puerarin respectively for 6 weeks.10 rats were selected to observe the pathological changes of periodontium and alveolar bone loss,serum of the other 10 rats were taken to determine AGEs,IL-6,TNF-α by ELISA. Results: The pathological changes of periodontium and alveolar bone loss of Puerarin group were obviously alleviated to compare with model group, the effects were similar to metformin group. Serum AGEs, IL-6 and TNF-α levels were levels of model group were higher than those in normal group(P<0.05);the three indexes were decreased significantly in Puerarin group and metformin group,there were statistic significance between the medicating groups with model group(P<0.05), the changes of Puerarin group were similar to metformin group.Conclusion:Puerarin Injection can effectively intervene rats’ diabetic periodontitis,reducing serum AGEs and lowering IL-6,TNF-αlevels.
