银屑病PBMCs和角质形成细胞对胃蛋白酶消化前后的β-溶血型链球菌的反应

摘要 目的 :探讨链球菌M蛋白在银屑病发病中的可能机制。方法:以胃蛋白酶消化前后的链球菌悬液作用于银屑病外周血单一核细胞 (PBMCs) ,用MTT方法检测细胞的增殖反应 ;应用3H -TdR掺入法检测链球菌对人角质形成细胞株Colo -16的作用。结果:银屑病患者PBMCs对胃蛋白酶消化后链球菌悬液的增殖反应明显下降 (P<0.05) ,然而 ,即使是消化后的链球菌也可以引起银屑病PBMCs增殖反应 (P<0.05) ,而正常对照组没有发现有统计学意义的增殖反应。胃蛋白酶消化前的链球菌可以抑制角质形成细胞DNA合成 ,消化后的链球菌对角质形成细胞没有明显的作用。结论:银屑病患者存在M蛋白反应性T淋巴细胞 。 Objective:To investigate the effects of streptococcal M protein on the pathogenesis of psoriasis.Methods:The responses of psoriatic peripheral blood mononuclear cells(PBMCs)to sonicated-streptococci in different concentration before(strep)and after(pep-strep)pepsinization were detected by means of MTT method.The effects of sonicated-streptococci in different concentration before and after pepsinization on keratinocytes were measured by3H-TdR incorporation assays.Results:The proliferative responses of psoriatic PBMCs to streptoˉcocci before pepsinization are greatly higher than that of pepsinated-streptococci(P<0.05,strep vs pep-strep).However,pepsinatd-streptococci could also improve the proliferation of PBMCs from psoriasis.As to the keratinocytes,sonicated-streptococci could greatly inhibit their3H-TdR incorporations(P<0.05,strep vs PRMI-1640).However,pepsinated-sonica_ ted-streptococci have no obvious effects on culture keratinocytes.Conclusion:There might be T-lymphocytes which specified to M protein in psoriatic patients.However,other antigens from streptococci might be implicated in the psoriatic pathogenesis.
