摘要
目的初步探讨肝气郁结证模型动物Th1、Th2细胞因子变化以及柴胡疏肝散对其干预作用的影响。方法应用RT—PCR法 ,检测肝气郁结证模型大鼠以及运用柴胡疏肝散、四君子汤治疗后T细胞IL—4、IFN—γmRNA的表达水平。结果与正常组比较 ,肝气郁结证模型大鼠IL—4mRNA表达较明显降低 (P <0 .0 5 ) ,IFN—γmRNA表达则明显升高 (P <0 .0 5 ) ,Th细胞Th1向偏移。用柴胡疏肝散治疗后 ,IL—4mRNA表达上调 ,IFN—γmRNA表达下调 ,与正常组相比较 ,基本恢复了正常水平 (P >0 .0 5 ) ,Th1、Th2趋于平衡状态 ;而四君子汤治疗后与正常组相比较 ,IFN—γ、IL—4mRNA的表达无明显变化 (P <0 .0 5 )。结论肝气郁结证模型大鼠Th1 /Th2细胞因子处于失衡状态 ,Th细胞向Th1偏移 ;柴胡疏肝散可以上调IL—4以及下调IFN—γmR NA表达 ,纠正肝气郁结证模型大鼠Th1 /Th2细胞因子的失衡状态。
Objective To investigate the expressive features of Th1/Th2 cytokines in rats with the pattern of Liver Qi stagnation(PLQS) and the intervening effect of Chaihu Shugan Powder(CSP) on it.Methods The expression of Th1/Th2 cytokines from T cells, including IL?-?4 and IFN-γ mRNAs,was determined by the method of RT-PCR in rats with PLQS,following the treatment with CSP and Sijunzi Decoction(SD) respectively.Results Compared with that of blank controlling group(BCG),the expression of IL?-?4 mRNA was significantly downregulated and that of IFN-γ mRNA was significantly upregulated in rats with PLQS(P< 0.05),suggesting a shifting of Th to Th1.When treatecl with CSP,these animals showed upregulated expression of IL?-?4 mRNA and downregulated expression of IFN-γ mRNA,with no significant difference between these and BCG (P>0.05).This meant a tendency to a balanced status of Th1/Th2 activity being established once more.In contrast,SD showed no significant effect on the expression of IFN-γ and IL?-?4 mRNAs.Conclusions The expression of Th1/Th2 cytokines is kept in an imbalanced situation among rats with PLQS, suggesting a shifting of Th to Th1 in their differentiation.On the other hand,CSP holds effects on the expression of these cytokines through upregulating the expressive activity of IL?-?4 and downregulating that of IFN-γ.In this way, the imbalanced situation of Th1/Th2 cytokine expression can be brought into a normal status.
