摘要 目的 :研究糖皮质激素对Ⅰ型超敏反应中的肥大细胞脱颗粒反应是否有直接作用。方法 :利用流式细胞仪技术观察地塞米松对RBL 2H3细胞 (大鼠嗜碱粒细胞 )脱颗粒程度的影响 ,利用westernblot分析ERK1 ERK2信号的改变。结果 :地塞米松预先作用于RBL 2H3细胞 ,使AnnexinV标记的细胞阳性率较致敏原活化组明显下降 ,MAPK活性明显降低。结论 :地塞米松抑制肥大细胞脱颗粒反应 。 AIM: To study the direct effect of dexamethasone on the degr an ulation of mast cells. METHODES: The flow cytometry was used to observe the effect of dexamethasone on the degranulation of RBL-2H3 cell line ( rat basophile leukemia cell line), and the changes of ERK1/ERK2 signal transduct ion characteristics was analyzed by western blot. RESULTS: Pre- treating RBL-2H3 cell line with dexamethasone resulted in the decrease of Annex in V positive staining of cells and the activity of MAPK compared that of the ac tivated group. CONCLUSION: The inhibition of dexamethasone on ma st cells is associated with the activity of MAPK.
