摘要
目的 研究去甲斑蝥素对裸鼠人胆囊癌移植瘤生长的抑制作用及其机制。方法 通过建立人原发性胆囊癌动物模型 ,应用去甲斑蝥素对裸鼠人胆囊癌移植瘤的增殖、生长进行药物对比抑制实验 ,观察、测定裸鼠移植瘤瘤体改变、抑瘤率和细胞凋亡率 ,并绘制生长曲线。结果 去甲斑蝥素的半数致死量 (LD5 0 )是 13 9.96mg·kg- 1,应用其 1/5LD5 0 剂量 ,即可使裸鼠移植瘤瘤体明显变小 (5.61± 0 .3 9vs. 9.78± 0 .61cm3 ,P <0 .0 5) ,抑瘤率达 42 .63 % ,瘤细胞凋亡率明显上升。
Objective To evaluate the inhibitory effects of norcantharidin (NCTD) on growth of implanted tumors of human gallbladder carcin oma in nude mice in vivo.Methods Animal model simulating human gallbladder carcino ma was established by subcutaneous implantation of cultured GBC-SD cell lines i nto nude mice.Mice used experiments were randomly divided into 4 groups and were taken different treatment.Tumor dimension was measured per 3 days.Tumor growth curve and inhibitory rate of each group were respectively evaluated.Flow cytom etry was used to measure the induction of apoptosis. Results LD50 of NCTD was 139.96 mg/kg.Volume of implanted tumors in nude mice in NCTD group (5.61±0.39 cm 3) was smaller than ones in c o ntrol group (9.78±0.61 cm 3, P <0.05).Tumor inhibitory rate in the NCTD grou p was 42.63%.Conclusion Norcantharidin can inhibit the growth and prol iferation of human gallbladder carcinoma cells in implanted tumor of nude mice.
