^(99)锝亚甲基二膦酸盐对Walker256癌荷瘤大鼠骨侵袭的抑制作用

Inhibitory Effects of ^(99)Tc-MDP Injection on Bone Invasion by Walker256 Carcinoma in Rats

目的 :研究99锝亚甲基二膦酸盐 (99Tc MDP)注射液对大鼠骨侵袭的抑制作用。方法 :建立Walker2 5 6癌荷瘤大鼠模型。分空白对照组、骨膦对照组及99Tc MDP 8,4,2 ,1mg 4个剂量组。腹腔内注射药物 10d后 ,处死大鼠取瘤组织称重 ,剥离左、右腿胫骨称重 ,摄胫骨X线片 ,作组织切片。结果 :与对照组比较 :99Tc MDP 8和 4mg组及骨膦对照组对减轻荷瘤侧胫骨增重及骨侵袭程度均差异有显著性 (P <0 .0 5 ) ;99Tc MDP 8和 4mg组与骨膦对照组相比差异无显著性(P >0 .0 5 ) ;99Tc MDP 2和 1mg组对癌细胞骨侵袭的抑制作用均差异无显著性 (P >0 .0 5 )。结论 :99Tc MDP及骨膦注射液对Walker2 5 6癌荷瘤大鼠均有抑制癌细胞骨侵袭的作用 ;在抑制程度上 ,骨膦注射液与99Tc MDP作用相当 ;99Tc MDP 4个剂量组的抑制程度与剂量呈正相关。

Objective:To investigate the inhibitory effects of 99 technetium methylenediphosphonate( 99 Tc-MDP) injection on bone invasion by walker 256 carcinoma in rats. Method:The experimental model of bone invasion by walker 256 carcinoma was made with 96 rats .The animals were divided into 6 groups, control group, 8 mg·kg -1 bonefos group, and 1, 2, 4, 8 mg·kg -1 99 Tc-MDP groups. The rats were injected in abdominal cavity with the drug for 10 days and then were killed on the eleventh day. The tibias' and tumor tissues' weights, roentgenogram and histology were examined. Results:The tibias' weight and the level of bone invasion by walker 256 carcinoma in 4, 8 mg·kg -1 99 Tc-MDP groups and bonefos group were significantly lower than those in the control group. The difference between the 99 Tc-MDP group and bonefos group was not significant. The effect of 1 and 2 mg·kg -1 99 Tc-MDP did not significantly differ from the control. Conclusion: 99 Tc-MDP and bonefos can inhibit bone 99 Tc-MDP increase with increasing doses.
