摘要
[目的 ]运用基因芯片技术 ,比较正常大鼠和糖尿病大鼠肝脏中细胞色素p45 0 2E1(CYP2E1)基因表达的差异。[方法 ]对雄大鼠腹腔注射链脲佐菌素 (STZ) ,并予以高脂饮食制造 2型糖尿病模型 ;从其肝脏抽提总RNA ,进行反转录 ,并用Cy3 dUTP和Cy5 dUTP分别对正常组与实验组探针进行荧光标记 ,探针与基因表达谱芯片杂交 ,结果由扫描仪扫描并用软件进行分析统计。 [结果 ]糖尿病组与对照组的肝脏相比 ,Cy5 /Cy3比值为 10 0 82 ,有统计学意义 ,CYP2E1mRNA表达升高。[结论 ]
