TGF-β_1和/或TNF-α反义PS-ODNS对造血干/祖细胞体外扩增的作用 被引量:3

Effects of TGF-β_1 and TNF-α antisense PS-ODNS on e x vivo expansion of hematopoietic stem/progenitor cells(HSPC)

摘要 目的 :研究TGF - β1和 /或TNF -α反义硫代寡核苷酸 (PS -ODNS)对造血干 /祖细胞体外扩增的调节作用。方法 :采用免疫磁珠分离技术从新鲜脐带血中分离CD3 4 + 细胞 ,用淋巴细胞分离液从骨髓血中分离单个核细胞 ,应用液体培养及造血祖细胞集落分析检测TGF - β1和 /或TNF -α反义PS -ODNS对CD3 4 + 细胞数及多向性造血祖细胞 (CFU -mix)、粒 -单祖细胞 (CFU -GM)、红系祖细胞 (BFU -E和CFU -E)集落数的影响。结果 :培养体系中加入TGF - β1反义PS -ODNS后CD3 4 + 细胞数、CFU -mix、CFU -GM、BFU -E和CFU -E集落数分别是对照组 (仅加细胞因子组 )的 4、2 6、2 7、1 8、2 1倍 ;加入TNF -α反义PS -ODNS后CD3 4 + 细胞数、CFU -mix、CFU -GM、BFU -E和CFU -E集落数分别是对照组的 4、2 9、2 6、1 7、1 8倍 ;同时加入TGF - β1和TNF -α反义PS -ODNS后CD3 4 + 细胞数、CFU -mix、CFU -GM、BFU -E和CFU -E集落数分别是对照组的 5 3、2 1、2 7、1 9、1 8倍。结论 :采用反义PS -ODNS阻断内源性TGF - β1和TNF -α是造血干 /祖细胞体外扩增的有效措施之一。 AIM: To study the effect of TGF-β 1 and TN F-α antisense PS-ODNS on ex vivo expansion of hematopoietic stem/progenitor cell s (HSPC). METHODS: CD 34 + cells were purified from fres h umbilical cord blood by immunomagnetic beads, and mononuclear cells were purifi ed from bone marrow by Ficoll-hypaque . The effects of TGF-β 1 and /or TNF-α an tisense PS-ODNS on ex vivo expansion of CD 34 + cells、CFU-GEMM、CFU-G M、CFU-E and BFU-E were detected by using liquid and semi-solid culture systems . RESULTS: TGF-β 1 antisense PS-ODNS cooperated with cytokines increased the number of CD 34 + cells,CFU-GEMM,CFU-GM,CFU-E and BFU-E , which was as 4,2.6,2.7,1.8,2.1 times as that of the control (the cytokine s combination), respectively. TNF-α antisense PS-ODNS cooperated with cytokines respectively increased the number of CD 34 + cells, CFU-GEMM, CFU-GM, CF U-E and BFU-E by 4, 2 9, 2 6, 1 7, 1 8 times as that of the control. The ab ove two antisense PS-ODNS cooperated with cytokines could respectively increased the number of CD 34 + cells, CFU-GEMM, CFU-GM, CFU-E and BFU-E by 5 3,2 1, 2 7, 1 9, 1 8 times as that of the control. CONCLUSION: I nhibition of endogenous TGF-β 1 and TNF-α by antisense PS-ODNS will be one of the effective methods to expand HSPC ex vivo.
