摘要
研究了葡萄糖限制的流加培养和批培养中杂交瘤细胞的生长、代谢和单抗生成。与批培养相比,流加培养的培养周期长,细胞密度、单抗浓度高。随着葡萄糖浓度的下降,葡萄糖比消耗速率和乳酸比生成速率也下降,葡萄糖转化成乳酸的比例从60%~70%下降到0,葡萄糖更多地参与三羧酸循环,提高了葡萄糖的有效利用率和ATP的生成效率。
The growth and metabolism and MAb production of hybridoma cells in a glucoselimited fedbatch culture and an ordinary batch culture were compared. In contrast to the batch culture, the culture cycle of fedbatch culture was prolonged, and cell density and MAb production were improved. The glucose concentration, the specific rates of glucose consumption and lactate production declined simultaneously, while the conversion rate of glucose to lactate decreased from 60%~70% to 0. It was shown that low glucose concentration caused the decrease of lactate accumulation facilating more glucose incorporated into TCA cycle, and high effective utilization of glucose and high production efficiency of ATP achieved.
