摘要
目的 血清对氧磷酯酶 1(PON1)的自动化检测方法建立及评价。方法 在Hitachi 76 0 0上建立自动化测定血清PON1的参数 ,对该方法的重复性、线性、干扰分析进行评估。并对正常人血清PON1活性进行分析。结果 低、中、高 3个浓度PON1的总变异分别为 6 .5 %、4 .2 %、1.7% ,测定线性可达 80 0U/ml。标本中胆红素浓度在 4 30 μmol/L以下 ,血红蛋白浓度在 5 .5 g/L以下以及三酰甘油浓度在 2 5 .0mmol/L以下对血清PON1的测定均无明显影响。正常人血清PON1活性呈正态分布 ,参考范围为 4 5 .5~ 2 6 8.8U/ml。结论 建立的血清PON1自动化检测方法简单、可靠、快速、廉价 ,适合临床实验室常规测定需要。
Objective To establish and evaluate an automatic analyzer method for detecting activity of paraoxo nase 1 (PON1) in serum. Methods The parameters for detecting human serum PON1 activity on Hitachi 7600 systems were established. The linearity, interference and reproducibility according to the items of NCCLS were evaluated. Results The total variations of low, medium and high levels samples were 6.5%, 4.2% and 1.7% respectively. This method for detecting the activity of PON1 in serum was not significantly interfered when the total bilirubin concentration was below 430 μmol/L, hemoglobin was below 5.5 g/L and triglyceride was below 25.0 mmol/L. The linearity was up to 800 U/ml. There was a normal distribution of PON1 in healthy population and the reference range was 45.5 268.8 U/ml. Conclusions The automatic analyzer method established is simple, reliable, and quick for detecting the activity of PON1 in serum.
