摘要
目的 探讨压缩空气反复暴露对外周血细胞含量的影响。方法 将大鼠置动物加压舱内 ,以 70 0 k Pa空气暴露 6 0 min,每天 2次 ,连续 3d。对照大鼠以 14 7k Pa纯氧或 70 0 k Pa含 2 1k Pa氧气的氮气处理。于暴露后第 1,3,5天晨断头取血。用自动生化分析仪测定外周血白细胞 (WBC)、淋巴细胞(L y)、粒细胞 (Gr)、红细胞 (RBC)、血红蛋白 (Hb)及血小板 (PL T)含量 ,用流式细胞仪测定外周血淋巴细胞亚型的变化。结果 高气压暴露后 ,外周血 WBC有升高倾向 ,PL T有降低倾向 ,但均无统计学意义(P>0 .0 5 )。 L y计数无变化 ,但其百分含量明显降低 (P<0 .0 5 ) ;Gr计数及百分含量均明显升高 (P<0 .0 5 ) ;CD3+ 、CD4 + CD3+ 细胞百分含量及 CD4 + CD3+ /CD8+ CD3+ 比值均显著下降 (P <0 .0 5 ) ,CD8+CD3+细胞含量变化无显著性 (P>0 .0 5 )。压缩空气反复暴露后第 3天 ,上述变化均恢复正常。RBC计数在暴露后第 1天轻微减少 ,但 Hb含量明显下降 (P<0 .0 5 ) ;第 3天时两者显著升高超过暴露前水平 (P<0 .0 1) ,至第 5天降至正常对照水平。 14 7k Pa纯氧处理后也出现上述变化 ,但常氧高氮处理后则无明显影响。结论 70 0 k Pa压缩空气反复暴露对大鼠血细胞计数有明显影响 ,表现有免疫抑制倾向 ,与呼吸气中高分压氧?
Objective To observe changes of peripheral blood cell count after repetitive air exposures.Methods Adult rats were exposed to 700 kPa air for 60 min in compression chamber 2 times a day for 3 consecutive days.Control rats were exposed to 147 kPa O\-2 or 700 kPa N\-2(containing 21 kPa O\-2).Peripheral blood were sampled after decapitation on the 1st,3rd and 5th day after exposures.The blood components including WBC,lymphocyte,granulocyte,RBC,hemoglobin and platelet were determined by biochemical auto-analyzer.The subsets of the lymphocyte were identified using 3-colour flow cytometry.Results After hyperbaric exposure,there existed an increasing inclination of WBC and a decreasing inclination of platelets but both were not statistically significant( P >0.05).The amount of lymphocyte didn′t change,but its percentage content decreased( P <0.05);the amount and the percentage content of granulocytes increased( P <0.05);the percentage content of CD3\++ and CD4\++CD3\++ cells and the CD4\++CD3\++/CD8\++CD3\++ ratio all decreased( P <0.05);the percentage content of CD8\++CD3\++ cells didn′t change( P >0.05).All these changes disappeared on the 3rd day after exposures.The RBC content decreased slightly while the hemoglobin content decreased significantly( P <0.05) on the 1st day after exposures,but both increased higher than normal on the 3rd day( P <0.01) and recovered to the control levels on the 5th day.Similar results appeared after 147 kPa O\-2 exposures but not after hyperbaric N\-2 exposures.Conclusions Peripheral blood component changes occurred after 700 kPa air repetitive exposures and exhibited an immunosuppressive inclination,which was related to the high partial pressure of O\-2 in the inspired gas of rats,but changes disappeared after 3~5 days.
