摘要
目的 观察 SD大鼠血管内皮细胞 (VEC)损伤后高压氧 (HBO)对内皮细胞修复中超微结构和功能的影响。方法 将 4 4只大鼠随机分成 6个组 :正常组 (N组 ,n=6 )、模型对照组 (M组 ,n=10 )、模型制作后 HBO治疗 5 d组 (T1 组 ,n=7)和 10 d组 (T2 组 ,n=7)以及高压空气 (HBA)治疗 5 d组 (T3组 ,n=7)和 10 d组 (T4 组 ,n=7)。血管内皮损伤模型均以皮下注射肾上腺素制成。观察其降主动脉VEC超微结构的改变 ,并用放免法测定血浆 ET- 1浓度变化。结果 T1 组和 T2 组血管内皮损伤均比 M组轻 ,血浆 ET- 1浓度也低于 M组 ,差异有显著性意义 (P<0 .0 5 ) ;T3组和 T4 组与 M组比 ,ET- 1浓度无明显变化。结论 HBO不但可以减轻 VEC损伤的发生发展过程 ,而且可以对损伤的血管内皮起到促进其修复和保护其功能的作用。
Objective To observe the effects of hyperbaric oxygen (HBO) on ultrastructure and function of injured vascular endothelial cell (VEC) in SD rats.Methods 44 rats were randomly divided into 6 groups:normal group(group N, n =6);model group (group M, n =10);hyperbaric oxygen treated model groups of 5 days (group T 1, n =7) and 10 days (group T 2, n =7);hyperbaric air treated model groups of 5 days (group T 3, n =7) and 10 days (group T 4, n =7). The rat model of injured VEC was established by injecting a high dose of adrenaline subcutaneously.The ultrastructure of VEC in the descending aorta and the change of ET-1 in plasma were observed.Results The extent of VEC injury in group T 1 and group T 2 were less severe than that in group M. The concentration of ET-1 in plasma in group T 1 and group T 2 were significantly lower than that in group M.The concentration of ET-1 in plasma in group T 3 and group T 4 were not significantly different from that in group M.Conclusions HBO may not only lessen the extent of VEC injury,but also improve the recovery of injured VEC and protect the function of injured VEC as well.
