摘要
目的 :观察丹参对H2 O2 诱导的离体大鼠主动脉平滑肌细胞 (VSMC)、增殖细胞核抗原 (PCNA)、核转录因子 (NF κB)蛋白表达和细胞增殖周期时相的影响 ,以探讨其可能的作用机制。方法 :以H2 O2 作为外源性活性氧(ROS)刺激VSMC增殖 ,用免疫细胞化学法检测培养的VSMC中PCNA和NF κB的表达 ,同时用流式细胞术测定细胞周期。结果 :H2 O2 可使VSMC胞核内PCNA和NF κB蛋白表达呈强阳性 ,并明显增加S期细胞百分比 (与对照组相比 ,P <0 0 5或P <0 0 1) ,加入丹参干预后 ,PCNA和NF κB蛋白表达明显受抑制 ,G0 /G1期细胞百分比升高 ,S期细胞百分比下降。结论 :抑制PCNA和NF κB蛋白表达 ,阻止细胞进入有丝分裂期可能是丹参抑制H2 O2 诱导的VSMC增殖的重要机制之一。
Objective: To observe the effect of Radix Salviae Miltiorrhizae Injection (RSMI) on proliferating cell nuclear angiten (PCNA), NF-κB and cell cycle phase of vascular smooth muscle cell(VSMC) induced by H 2O 2 in vitro. Methods: H 2O 2 was added to the medium for culturing rat aortic VSMC as extraneous reactive oxygen species (ROS). The protein expression of PCNA and NF-κB was examined with immunocytochemical technique in VSMC, and cell cycle phase analysis was used to detect proliferation of VSMC. Results: The result showed that the protein expression of PCNA and NF-κB became stonger, the ratio of S phase cells became higher in H 2O 2 group. RSMI could increase the percentage of the cells in G 0/G 1 phase, decrease the percentage of the cells in S phase and obviously inhibit the expression of PCNA and NF-κB proteins. Conclusions: The expression of PCNA and NF-κB was inhibited and the transition from G 0 phase to S phase was blocked, which may be one of the key mechanism of RSMI on the inhibition of VSMC proliferation induced by H 2O 2.
