养心通脉片对胰岛素抵抗综合征外周血单核细胞C-myc和PDGF-A mRNA表达的影响

Effects of YTT on mRNA Expression of C-myc and PDGF-A in the Patients with IRS

目的 :探讨养心通脉片防治胰岛素抵抗综合征 (IRS)的分子生物学机制 ,主要是检测分析该药对IRS病人外周血单核细胞C -myc基因和血小板衍化生长因子 -A (PDGF -A )mRNA表达水平的影响。方法 :设养心通脉片组( 3 3例 )、二甲双胍片组 ( 3 4例 )。两组病例分别于服药前、服药后的第 10天采空腹血液 ,用常规方法定量检测血糖(FPG) ;用放射免疫法检测血胰岛素(FINS) ;用半定量RT -PCR法检测外周血单核细胞C -myc基因和PDGF -AmRNA的表达水平。结果 :养心通脉片组病人服药后FPG、FINS含量与服药前比较明显下降 (P <0 .0 1) ,胰岛素敏感指数 (ISI)与服药前比较明显上升(P <0 .0 1) ;养心通脉片组病人服药后外周血单核细胞C -myc基因、PDGF -AmRNA的表达水平与服药前比较明显下降 (P <0 .0 1) ,其中C -myc基因mRNA表达下降的幅度大于二甲双胍片组 (P <0 .0 1)。结论 :养心通脉片能通过改善机体胰岛素敏感性 ,抑制C -myc基因mRNA的表达 ,而下调PDGF -AmRNA的表达水平 ,有利于进一步抑制血管平滑肌细胞的过度增殖 ,防止IRS的发生、发展。

Objective:To explore the molecular mechanism of Yangxin Tongmai Tablet (YTT) in preventive and curative insulin resistance syndrome (IRS) through checking the effects of YTT on mRNA of C-myc and platelet derived growth factor-A (PDGF-A) in unit-nuclear cell of peripheral circulation. Methods:77 cases with IRS patients were randomly divided into 2 groups, viz. 33 and 44 cases treated with YTT and metformin (met), respectively. Fasting plasma glucose (FPG) and insulin(FINS), etc. were measured by the conventional method, while the expression level of mRNA of C-myc and PDGF-A was tested with the means of reverse transcriptase-polymerase chain reaction (RT-PCR). Results:The contents of FPG and FINS after treatment showed significantly dropper (P<0.01), while insulin sensitivity index (ISI) was higher and the expression level of mRNA of C-myc and PDGF-A was lower than before treatment (P<0.01). However, compared with met group, the level of c-myc mRNA expression in YTT group was remarkably reduced (P<0.01). Conclusion:YTT had the roles with inhibiting over proliferation of vascular smooth muscle cells and preventing formation and development of IRS through improving insulin sensitivity, inhibiting C-myc mRNA expression and following reducing the level of PDGF-A mRNA.
