龟板对脊髓损伤后大鼠功能和骨形态发生蛋白4表达的影响 被引量:8

摘要 目的 观察龟板对大鼠损伤脊髓骨形态发生蛋白 4 (BMP4 )表达和后肢功能恢复的影响。方法 应用改良Allen脊髓损伤模型 ,于术后 1、7、14、2 1、2 8d分别对大鼠进行Tarlov评分和斜板试验检查后肢功能。应用免疫组织化学技术检测BMP4的表达 ,观察龟板对脊髓损伤后BMP4的影响。结果 脊髓损伤后第 1d ,在两组受损伤脊髓灰质中都可见到BMP4的表达。第 14d时达到高峰 ,龟板组为 37 2 4± 5 73,损伤组 2 1 4 8± 6 83(P <0 0 5 )。龟板组可使损伤的脊髓BMP4持续高表达至术后第 2 8d ,而损伤组仅持续表达至术后 2 1d。增加的BMP4阳性细胞数与神经功能的改善平行。 Objective To investigate the effect of Tortoises shell on expression of BMP4 in spinal cord following injured spinal cord as well as functional recovery in the rats. Methods Sixty six adult SD rats were randomly divided into two groups: Tortoises shell therapy group,spinal cord injury group. After operation open field and inclined plane tests were performed to test the function of the rear limbs. The animals were sacrificed 1,7,14,21 and 28 day after surgery. Detect expression of the BMP4 by immunohistochemical method, the effects of Tortoises shell on expression of BMP4 in spinal cord following injured spinal cord were observed. Results The BMP4positive expression cells were detected in the gray matter one day after spinal cord injury(SCI) The BMP4 positive expression cells increased after operation and reached their peak at day 14 after SCI in both group; The number of the BMP4 positive expression cells in spinal cord injury group and in Tortoises shell therapy group was 21 48±6 83,37 24±5 73 respectively( P <0 05). The positive cells in Tortoises shell therapy group maintained at high level to day 21 ,however, that in spinal cord injury group decreased significantly at 21 day. The increase in the BMP4 positive cells was paralleled by a significant improvement in neurological function evaluated by open field and inclined plane test. Conclusion Tortoises shell can alleviate the neurological symptoms and promote the expression of BMP4 in spinal cord following spinal cord injury of rats.
