摘要 目的 为了初步了解胰岛素增敏剂罗格列酮对自发性高血压大鼠的降压作用和机理 ,设计了本项实验。方法自发性高血压大鼠 3 6只 ,随机分为 3组 ,罗格列酮和硝苯吡啶分别作为观察药和阳性对照药添加到大鼠饮水中 ,另有一组空白对照。结果  2种药物均呈现显著的降压作用 ,但血胰岛素水平测量结果没有达到显著的统计学差异。结论 胰岛素增敏剂 -罗格列酮可以显著降低自发性高血压大鼠的血压 ,其机理有待深入探讨。 Objective To evaluate the effect of insulin sensitized agent (rosiglitazone), on blood pressure in spontaneous hypertensive rats (SHR). Methods Thirty six SHR were randomly assigned to receive drinking water with nifedipine ( n =12, 140 mg/L) or rosiglitazone ( n =12, 10 mg/L in drinking water) for 2 months. Twelve SHRs received drinking water as control. BP was measured by tail cuff methods and plasma insulin was determined by radio immunoassay. Results BP was decreased in both rosiglitazone and nifedipine groups starting at the 1~2 weeks of treatment (rosigitazone group 147 mmHg, nifedipine, 132 mmHg vs controls 174 mmHg). The differences of BP between the treated and untreated groups were sustained until the end of 8 th weeks of treatment (rosiglitazone group:SBP 162 mmHg, nifedipine, 161 mmHg, control 184 mmHg). Plasma insulin were decreased in both treated groups but not reached significance. Conclusion Insulin sensitized agent, rosiglitazone, significantly decreased SBP in SHRs. The mechanism by which the reduction of BP needs further investigation.
