摘要 目的:从细胞分子水平研究抗纤软肝冲剂对肝星状细胞(HSC)表达转化生长因子β1(TGF-β1)的影响。方法:抗纤软肝冲剂药物血清温育传一代HSC,以生理盐水和秋水仙碱为对照。免疫细胞化学分析TGF-β1 的蛋白合成;半定量RT-PCR 法检测TGF-β1 的mRNA 表达量。结果:抗纤软肝冲剂能明显抑制HSC 的自分泌TGF-β1及其mRNA 表达(P<0.01)。结论:抗纤软肝冲剂能抑制HSC 的TGF-β1 的基因和蛋白表达,阻断其自分泌放大活化效应,这可能是该方抗肝纤维化的细胞分子学机制之一。 Objective:To study the effect of Kangxian Ruangan Granule (KXRGG) on the expression of TGF-β1 at the cellular and molecular level.Methods :The medicated sera were prepared to incubate with the subcultured HSC, sodium chloride sera and colchicines sera were taken as control groups. TGF-β1 protein production was examined by immunocytochemistry stain ;the gene expression of TGF-β1 was analyzed by semiquantitative RT-PCR. Result :KXRGG obviously inhibited the expression of TGF-β1 mRNA and its self-secretion. Conclusions:KXRGG can inhibit the gene expression and protein secretion of TGF-β1 of HSC, and obstruct its amplified auto-activation, which may be the cellular and molecular mechanism of its antifibrotic action.
