摘要 尼群地平(nitrendipine)为一新的二氢吡啶类钙离子通道阻断药。其抗高血压作用较硝苯吡啶强且持久,尤其适用于老年患者。该药的有效血浓度较低,故文献中多采用高灵敏的检测方法:电子捕获鉴定器(ECD)及色谱-质谱联用(GC-MS)。 A specific and sensitive method has been established for analysis of nitrendipine in human plasma by gas chromatography with a mass selected detector. Nitrendipine and diazepam (internal standard) were extracted from plasma with 3. 0 ml of diethyl ether- cyclohexane(2 : 1) after the addition of 0. 1 ml NaOH. The organic phase was then evaporated with a mild nitrogen stream and the residue was redissolved in 20 ul of ethyl acetate, of which 1 - 2 ul was injected into a fused-silica capillary column (12. 5m × 0. 2mm. cross-linked dimethyl silicone) equipped in an HP5970B GC-MSD. Mass 238 for nitrendipine and 256 for diazepam were quantitatively measured by selected ion monitoring.The area ratio (nitrendipine/diazepam) was linear to the concentration of nitrendipine over the range of 0-61. 08ng/ml(γ=0. 9999). The coefficient of variation of intra and inter-day assay was less than 5 % and 8 %, respectively. The average recovery from spiked plasma was 97. 30%. The method has been applied in clinical analvsis.
