摘要
应用聚丙烯酰胺凝胶电泳技术,对引自国际干旱农业研究中心的4个春箭舌豌豆品系原种(F0)和在甘肃省甘南藏族自治州经过3个世代选育的F3代,进行幼苗下胚轴过氧化物酶同工酶分析。结果表明,2505品系F3代在相对迁移率(Rf)为0.725位置和原种相比酶带缺失,而Rf值为其它位置处谱带颜色均较原种为浅;2556F3代在Rf值为0.725处呈现酶带,而原种该谱带缺失,其它位置各谱带明显较原种为深;2560F3代在Rf值为0.211和0.266位置处酶带变宽,颜色加深;2566F3代在Rf值为0.725处呈现酶带而原种该谱带缺失,在Rf值为0.211和0.266处2酶带较原种位置偏上,颜色偏深。供试8个品系过氧化物酶同工酶酶谱与对照品种333/A相比,均存在较大差异。由此对以上4品系原种和F3代呈现出的宏观上的生态适应性提供了分子水平上遗传变异依据。
Four lines of Vicia sativa were introduced from the International Center for Agricultural Research in Dry Areas (ICARDA).The codes of the introduced lines were 2505, 2556, 2560 and 2566. They were evaluated and selected for adaptation to local environments, and high seed and herbage yields, under alpine grassland conditions at Xiahe County in the Gansu province of China. Performance of F3was significantly better than its parent generation. Therefore, seedling hypocotyl POD isozyme PAGE was used to study the genetic mechanism of F0 and F3of the 4 lines tested .The results showed that the mapping of POD isozyme PAGE was markedly different between F0and F3. For the line 2505, F3missed one band at the loci Rf 0.725 when compared to F0, and in other locations the bands of F3were lighter than those of F0. For the line 2556, F3presented one band at the loci of Rf 0.725 whereas this was missing for F0, and at other locations, the bands of F3were darker than those of F0. For the line 2560, the two bands at the loci Rf 0.211 and 0.266 of F3became wider and darker than those of F0. For the line 2566, F3presented one band at the loci Rf 0.725 which was missing in F0, and at other locations the bands of F3were darker than those of F0. 333/A (CK) POD isozyme mapping showed marked differences to tested lines. The study suggests that POD isozyme PAGE offers evidence of the ecological adaptability of F3of 4 lines of V.sativa at the molecular level.
