人AHH1 T淋巴细胞辐射死亡与凋亡的关系及其意义

摘要 应用原位末端标记(TUNEL)和麦格-姬姆萨(MGG)染色和MTF比色法研究辐射诱导的人AHH-IT淋巴细胞死亡与凋亡的关系、细胞活存及其意义。结果表明,经2~10 Gy γ-线照射后4~48 h,随着照射剂量的增加和照后时间的延长,淋巴细胞凋亡指数明显升高,显示出较好的剂量和时间效应关系;而相同剂量照射后,淋巴细胞活存率随照射剂量增加而明显减少。上述结果表明,在一定照射剂量范围内,凋亡是辐射诱导淋巴细胞死亡的重要原因之一。 By using TUNEL and MGG staining methods and MTT colorimetric analysis, the relationship of the death and apoptosis of radiation-induced human AHH1 T lymphocyte, cell survival and their significance were investigated. The results show that apoptotic index of lymphocyte increased obviously with the increment of radiation doses and time prolongation after radiation 4~48 h after 2~10 Gy γ-irradiation, indicating a good relationship of dose-effect, while the lymphocyte survival rate decreased significantly with the elevate of radiation dose, showing a negative relations. Above-mentioned results suggested that in a definite range of radiation doses, apoptosis was one of the major causes of radiation-induced lymphocyte death, which provides an important experimental basis to prevent ant treatment immune damage of acute radiation sickness from a point of inhibiting lymphocyte apoptosis.
