氯化镧对创口组织中不同型胶原蛋白表达的影响 被引量:1

摘要 目的 了解氯化镧对Ⅰ、Ⅱ、Ⅲ和Ⅳ型胶原蛋白表达的影响和防治瘢痕的可能性。方法 以SD大鼠为模型 ,在背部做切口 ,处理方法分为 4种 :空白对照组 (切口两侧皮下不注入任何液体 )、模拟对照组 (注入药物溶剂三蒸水 )、氯化镧组 (5 0mmol/L 0 .2 5ml)和抗TGF - β1抗体组 (5 0 μg/切口 )。在 14、2 8天后 ,用亲合素 -生物素 -过氧化物酶技术 (ABC法 )对伤口组织进行各型胶原水平的检测。结果 应用氯化镧后 ,Ⅰ、Ⅲ和Ⅳ型胶原蛋白的表达均显著降低 (P <0 .0 5 ) ,与使用抗TGF - β1抗体的作用相似。结论 氯化镧可以抑制创口组织中胶原蛋白的表达 ,有一定的防治瘢痕的作用 ,值得进一步研究和探索。 Objective In order to understand the effect of lanthanum chloride on expressions of type Ⅰ, Ⅱ, Ⅲ and Ⅳ collagen proteins and the possibility to prevent and treat scar.Methods Linear incisions were performed on dorsal skin of adult female Sprague\|Dawley rats. All rats were divided into 4 groups according to different management of wound: wounds in the control group were treated with nothing; wounds in the sham\|control group were injected with distilled; wounds in the experiment group were injected with 50 mmol/L of lanthanum chloride; wounds in the positive control group were injected with 50μg neutralizing antibody to TGF\|β1. Volume of different solutions injected on each wound was 0.25 ml. On the 15th and 29th day after incision, the wound tissues were harvested and levels of various types of collagens were assayed by immunohistochemistry (ABC Method).Results In the experimental group, the expressions of typeⅠ、Ⅲand Ⅳ collagen proteins reduced significantly (\%P\%<0.05). The effect of lanthanum chloride on expressions of type Ⅰ, Ⅲ and Ⅳ collagens was similar to that of neutralizing antibody to TGF\|β1.Conclusion Lanthanum chloride can certainly prevent and treat scar by inhibiting expressions of collagen proteins and is worthy of studying and exploring further.
