人子宫内膜异位症小白鼠模型的建立及雌、孕激素受体的表达(英文) 被引量:4

ESTABLISHMENT OF ENDOMETRIOSIS MOUSE MODEL AND EXPRESSION OF STEROID HORMONE RECEPTORS

摘要 目的 为子宫内膜异位症 (EM)的临床研究提供实验动物模型。方法 选择成熟雌性小白鼠 ,切除卵巢。实验组 :取EM患者和正常育龄妇女分泌晚期子宫内膜 ,剪碎 ,随机置入小白鼠盆腹腔 ;对照组 :同法置入人大网膜。实验Ⅰ组和对照组给雌激素支持 ,实验Ⅱ组不给雌激素。各组小白鼠分别于 5、10、15、2 0d脱颈椎处死 ,观察异位病灶生长情况。免疫组化法检测异位病灶雌、孕激素受体表达。结果 子宫内膜种植第 5天 ,腹膜等部位即见异位病灶生长 ,可见腺体细胞及散在间质细胞 ,时间越长 ,病灶与小白鼠组织融合越明显 ,粘连越重。用EM患者和正常育龄妇女内膜种植的异位病灶无差别 ,实验Ⅰ组和Ⅱ组所形成的异位病灶亦无差别 ,它们的雌、孕激素受体表达未显示差别 ,多数呈弱表达。结论 小白鼠做为EM早期动物模型科学、简便、可行 ,雌激素对早期异位病灶发生无影响 ,EM是与多因素相关的全身性疾病。 Objective:To establish an experimental endometriosis model using mouse as a xenographic host for clinical study of endometriosis.Methods:Mature female mice were all ovariectomied. Mice of experimental groups were implanted with late secretory endometrium from normal people and patients with endometriosis. Mice of experimental group 1 were given estradiol support and experimental group 2 were given saline support. Mice of control group were implanted with human greater omentum given estradiol support. Mice were killed at 5 d, 10 d, 15 d and 20 d after transplantation to observe the growth of ectopic lesions. Meanwhile, we detected the expression of steroid hormone receptors by immunohistochemical method.Results:Ectopic growth of endometrial fragments was observed on the 5th day after peritoneal implantation, which revealed endometrial gland and stroma in ectopic lesions. And the adhension was more and more severe with the enlogation of time. There were differences of ectopic lesions neither between the implantation by normal endometrium and endometrium from patients with endometriosis, nor between experimental group 1 and experimental group 2. lmmunochemistry also revealed no differences and almost weak expression of steroid hormone receptors.Conclusions:The mouse is an appropriate model for the study of early phase of endometriosis. There are no striking influences of oestrogen and local factors of endometrium in the early phase of endometriosis.
