摘要
目的建立HPLC法测定甘草附子汤中甘草苷和香豆素含量的方法。方法采用HypersilC18色谱柱 (2 0 0mm× 4 6mm ,5 μm) ,流动相为甲醇 乙腈 水 冰醋酸 (体积比 5∶1 5∶80∶1 ) ,流速0 8mL·min-1,检测波长 2 5 4nm。结果甘草苷在 2 0~ 3 0 0mg·L-1内成良好线性关系 ,平均回收率 95 9% ,RSD为 3 1 %。香豆素在 1 7~ 1 6 7mg·L-1内成良好线性关系 ,平均回收率 95 9% ,RSD为 2 0 %。
Objective An HPLC method was developed to determine the contents of liquiritin and coumarin in the Gancao-Fuzi Decoction.Methods The separation was performed on Hypersil C 18 column ( 200 mm× 4.6 mm, 5 μm) with methanol-acetonitrile-water-acetic acid (5∶15∶80∶1) as the mobile phase.The flow rate was 0.8 mL·min -1 and the detection wavelength was 254 nm.Results The standard curve of liquiritin was linear in the range of 20~ 300 mg·L -1. The average recovery was 95.9% with RSD 3.1%.The standard curve of coumarin was linear in the range of 1.7~ 16.7 mg·L -1. The average recovery was 95.4% with RSD 2.1%.Conclusions The method is simple, rapid and accurate, it can be used for the quality control of the Gancao-Fuzi Decoction.
