摘要
目的 研究创伤感染时小鼠肝组织中锌指蛋白A2 0的表达规律及其对内毒素性肝损害病理改变的影响。方法 选用健康昆明种小白鼠 95只 ,雌雄不拘 ,体重 18~ 2 2 g ,制作双侧股骨闭合性骨折创伤及创伤合并内毒素血症模型。随机分为正常对照组、单纯创伤组、单纯内毒素组和创伤内毒素组。以Westernblot技术检测肝组织中锌指蛋白A2 0的含量变化 ,结果以 5次实验结果的A2 0蛋白条带平均吸光度值的平均值作为蛋白含量的相对值。结果 正常对照组A2 0蛋白呈现低量表达 (12 .90± 2 .18) ;单纯创伤组各时相点与正常对照组相比差异无显著性 ;单纯注射内毒素后 0 .5h ,A2 0蛋白含量较对照组升高明显 (2 3 .80± 4.61,P <0 .0 5 ) ,至 2 .0h表达又有所升高 (2 8.40± 10 .2 0 ) ,内毒素注射后 4.0~ 7.0h为A2 0蛋白表达的高峰期 ,且较单纯创伤组显著升高 ;而创伤并注射内毒素后 0 .5hA2 0蛋白表达较单纯创伤组 0 .5h显著增高 (3 4.5 3± 1.64 ,P <0 .0 1) ;至 2 .0h表达又明显升高 (4 1.94± 3 .0 7) ,且也显著高于单纯创伤 2 .0h(P <0 .0 1) ;2 .0h后A2 0蛋白表达仍呈升高趋势 ;创伤并注射内毒素后 7.0h时表达至高峰 (5 4.2 0± 5 .61) ,与单纯创伤组相比差异有显著性 (P <0 .0 1)。病理观察结果显示 :创伤并注射?
Objective To study the expression and characteristics of zinc finger protein A20 of mouse liver tissues in the process of trauma with infection and the effect on the posttraumatic secondary endotoxic liver lesions.Methods The models of fracture of trauma with infection were made.Ninty five healthy mouse of both sexes with a mean body weight of 20 g (18~22 g) were randomized into 4 groups:control,trauma,LPS and trauma plus LPS.The expression and characteristics of zinc finger protein A20 of liver tissues was measured by using western blot.All data were expressed as optical density of A20 protein strap.Results In control group,the A20 protein was expressed low (12.90±2.18),and there was no significant difference in the expression of A20 protein between traum group and control group.The expression of A20 protein was elevated to (23.80±4.61) at 0.5 h after injection of LPS,which was significantly higher than in control group ( P <0.05),and advanced to (28.40±10.20) at 2.0 h. The expression of A20 reached a peak during 4.0~7.0 h,and decreased after 7.0 h.However, 0.5 h after trauma and injection of LPS,the expression of A20 protein was elevated to (34.53±1.64),which was significantly higher than that of 0.5 h in trauma group ( P <0.01),and further to (41.94±3.07) at 2.0 h.The expression of A20 protein remained and increased tendency after 2.0 h.The expression of A20 protein reached summit (54.20±5.61) at 7.0 h,significantly higher than in trauma group ( P <0.01).Then the expression of A20 protein showed a descent tendency at 10.0 h.Pathologic observation showed that after 0.5 h~4.0 h of trauma and injection of LPS,serious pathologic lesions occurred in the liver lesions,alleviated in 7.0 h,but severer than in control group.Conclusion zinc finger protein A20 is highly expressed as an endogenous anti inflammatory protein in the earlier period of trauma with infection.It further indicates that temporarily increased expression of A20 in tissues following traumatic infection might play an important role in prevention of uncontrolled inflammatory response in liver.
