肺癌中p53和p63蛋白表达的高通量组织微阵研究 被引量:1

摘要 目的 探讨p5 3和 p6 3蛋白在肺癌中的表达及意义 ,并比较两者之间的表达关系。 方法 组织微阵 (tissuemicroarray ,TMA)技术和免疫组化S P方法 (immunohistochemistry ,IHC)。 结果 p5 3蛋白IHC染色中 ,TMA有价值的肿瘤标本为 2 34(72 .9% ) ,总的表达率 (+、++、+++)率为 4 8.3% (113/ 2 34)。随着肺鳞状细胞癌 (squamouscellcarcinoma,SCC)和腺癌(adenocarcinoma ,Ad)分级升高 ,p5 3蛋白表达增强 ,与肺SCC及Ad病理分级有明显相关性 (P <0 .0 5 )。p5 3蛋白表达与生存期呈明显负相关性 (P <0 .0 5 )。p5 3蛋白表达与病理分型和临床分期等无明显相关性 (P >0 .0 5 )。p5 3蛋白表达在肺癌原发肿瘤和转移淋巴结之间无明显差异 (P >0 .0 5 )。p6 3蛋白IHC染色中 ,TMA有价值的肿瘤标本为 2 6 4 (82 .2 % ) ,总的表达率(+、++、+++)为 6 7.8% (179/ 2 6 4 )。p6 3蛋白在小细胞肺癌 (smallcelllungcarcinoma,SCLC)表达最低 16 .0 % (4 / 2 5 ) ,非小细胞肺癌 (non smallcelllungcarcinoma ,NSCLC)与SCLC之间表达存在明显差异 (P <0 .0 5 )。p6 3蛋白表达与病理组织分级、临床分期、生存期等无明显相关性 (P >0 .0 5 )。p6 3蛋白表达在肺癌原发肿瘤和转移淋巴结之间无明显差异 (P >0 .0 5 )。结论  1.p5 3? Objective To value abnormal expression of p53 and p63 protein in order to reveal their potential relevance to diagnosis and prognosis and to compare p53 and p63 expression in lung cancers. Methods Immunohistochemical staining for p53 and p63 were carried out with S-P method in tissue microarray(TMA). Results In the TMA blocks there were 234(72.9%) cancer specimens valid for p53 protein assessment. A total expression rate was 48.3%(113/234). p53 protein expression was significantly associated with histopathologic grading and the survival rate(P<0.05). However, the expression of p53 protein was not associated with histopathologic primary cancers and metastasis lymphnodes. In the TMA blocks there were 264 (82.2%) cancer specimens valid for p63 protein assessment. A total expression rate was 67.8%(179/264). p63 protein expression in NSCLC was significantly stronger than that in SCLC (P<0.05). However, the expression of p63 protein was not associated with clinical staging 、histopathologic grading and survival time. The expression of p63 was not significantly different between primary cancers and metastasis lymphnodes. Conclusion The studies showed that: 1. p53 is an independent prognostic marker in lung cancers; 2.The expressions of p63 protein are different in different subtypes of lung cancers. It may indicate different molecular mechanism in different subtypes; 3. There are no correlations observed between the expression of p53 and p63; 4. This study shows that tissue microarrays can be used effectively for evaluation of the expression of certain tumor markers.
