DNA高甲基化与抑癌基因 被引量:9

摘要 哺乳动物基因组中,DNA甲基化是指 CpG二核苷酸中的胞嘧啶第5位碳原子被甲基化.DNA甲基化是一种基因外修饰,不改变DNA的一级结构;他在细胞正常发育、基因表达模式以及基因组稳定性中起着至关重要的作用。全基因组低甲基化,维持甲基化模式酶的调节失控和正常非甲基化CpG岛的高甲基化是人类肿瘤中普遍存在的现象。DNA高甲基化是导致抑癌基因失活的又一个机制.本文综述了抑癌基因的高甲基化、DNA修复基因的高甲基化、甲基化与转录的关系以及导致转录失活可能存在的作用机制、寻找甲基化相关基因的依据原则、甲基化的检测方法、肿瘤甲基化图谱的特征、甲基化与突变的相互作用、导致甲基化产生的原因、及其广泛的应用前景.
