昆明山海棠雄性抗生育活性提取物TH5的毒性评价 被引量:2

摘要 目的:对昆明山海棠雄性抗生育活性提取物TH5的毒性试验结果进行评价。方法:TH5的毒性试验用急性毒性试验(LD50)、Ames试验、CHL细胞染色体畸变试验及微核试验方法进行检测。结果:TH5之ID_(50)为3.27g/kg;Ames试验在0.5~5000μg/皿5个剂量范围内加与不加S9条件下,4个菌株的回变菌落数均未有2倍增加;TH5之CHL细胞染色体畸变试验在其最终浓度为1/4IC50、1/2IC50及IC50(11~44μg/ml)时无论加S9与否,细胞畸变率与溶剂对照组无显著差异(P>0.05);TH5在30~300mg/kg3个剂量水平其微核率与溶剂对照组也均无显著差异(P>0.05)。结论;TH5的抗生育有效剂量(116mg/kg)范围内3种诱变试验结果均为阴性,仍不失其开发价值。 Objective: To appraise the toxicity of TH5 of the male antifertility activity extracts in Tripierygi-um Hypoglaucian ( Levees ) Hutch. Methods: The toxicity of TH5 was examined using LD50, Ames test, CHL cell chromosome aberration lest and micronuclei test. Results: LD50 of 0.5 - 5000μg/plate did not increase the number of reversion mutation ( < 2) in any one of the four stains with S9 or without S9 in Ames test. In CHL cell chromosome aberration test. the cell leratogenesis ration of TH5 was no significant difference compared with the solvent control group (P >0. 05). Concentration ranged from 1/4 IC50 to IC50( 11 - 44 ) (μg/ml, + S9 or -S9) . The frequencies of micronucleus cell of TH3 in 3groups(30.150,300mg/kg) were no significant difference compared with the solvent control group (P >0. 05) . Conclusion; The mutage-nicity of TH5 in the antifertility active dose( 116mg/kg) were negative.
