摘要
目的 探讨电离辐射对BALB/c小肠上皮细胞PrxⅠ和PrxⅥ基因表达的影响。方法 采用60 Co辐射源全身照射BALB/c小鼠的方法建立电离辐射损伤的动物模型 ,总剂量为 9.0Gy ;在照射前 ,照射后 3、2 4h和 72h分离BALB/c小鼠的小肠上皮细胞 ;采用Tripure试剂盒提取小肠上皮细胞总RNA ,而后采用逆转录聚合酶链式反应 (RT PCR)的方法检测PrxⅠ和PrxⅥ基因表达 ,并进行半定量分析。结果 成功的分离了小鼠小肠上皮细胞 ;采用Tripure试剂盒提取的小肠上皮细胞总RNA经OD2 60 /OD2 80 比值测定和甲醛变性凝胶电泳证实所提取的RNA无降解并有足够的程度 ;正常小肠上皮细胞内有PrxⅠ和PrxⅥ基因的表达 ,在电离辐射后两者均有一短暂降低 ,随后逐渐升高 ,至照射后 3d仍高于正常对照。结论 电离辐射后PrxⅠ和PrxⅥ基因表达在应激期内出现一短暂性降低 ,在辐射后期PrxⅠ和PrxⅥ基因表达升高 ,提示PrxⅠ和PrxⅥ基因可能参与辐射引起的小肠上皮细胞修复。
Objective To explore the effect of ionizing radiation on the expression of PrxⅠand PrxⅥ in murine intestinal epithelial cells. Methods Animal model of ionizing radiation induced damage was established with the use of 60 Co radiation source, 9.0 Gy of total dose. Murine intestinal epithelial cells were isolated from BLAB/c mice on 0 h, 3 h, 24h, and 72 h after ionizing radiation, respectively. PrxⅠand PrxⅥ gene expression in murine intestinal epithelial cells were semi quantitatively with reverse transcription polymerase reaction (RT PCR), followed by isolating total RNA with Tripure kit. Results Murine intestinal epithelial cells were isolated successfully. RNA, isolated by Tripure kit, was proved as high purity and integrity. There are expression of PrxⅠand PrxⅥ in murine intestinal epithelial cells from untreated mice. The expressions of PrxⅠand PrxⅥ were transiently down regulated by ionizing radiation, however, then up regulated in the following time. Conclusion Our present results suggest that PrxⅠand PrxⅥ gene are involved in the ionization radiation induced damage and the repair of intestinal epithelial cells.
