丹参酮ⅡA抑制多种肿瘤细胞生长的体外实验研究

The inhibiting effect of TanshinoneⅡA on the growth of tumor cell lines in vitro

目的 研究丹参酮ⅡA对多种不同类型肿瘤细胞生长的体外抑制作用。方法 用 0 .5 μg·ml- 1 丹参酮ⅡA处理体外培养的肿瘤细胞 4d ,分别应用光镜、细胞计数、集落形成试验、流式细胞仪分析丹参酮ⅡA对不同肿瘤细胞生长的影响。结果 经丹参酮ⅡA处理后 ,肿瘤细胞生长和增殖明显被抑制 ,对NB4细胞的生长抑制作用更明显 ;部分肿瘤细胞发生凋亡的细胞形态改变 ,凋亡百分率为 10 .0 %～ 2 4 .3% (对照组为 2 .1%～ 6 .9% ) ,细胞明显被阻止于G0 G1 期 ,而S期细胞明显减少。结论 0 .5 μg·ml- 1 丹参酮ⅡA可明显抑制多种肿瘤细胞生长 。

OBJECTIVE To study the inhibiting effect of TanshinoneⅡA (Tan ⅡA)on the growth of various tumor cell lines. METHODS The tumor cell lines cultured in vitro were treated with 0.5 μg·ml -1 Tan ⅡA for 4 d. The morphology of the cells were observed by microscopy. The cell growth and proliferation were measured by cell counting, plating information and flow cytometry.RESULTS 0.5 μg·ml -1 Tan ⅡA could obviously inhibit the growth and proliferation of tumor cell lines(the inhibiting effect was more obvious on NB4 cell line). The morphologic change of apoptotic cells were found 10.0%-24.3% in part of the tumor(the control group are 2.1%-6.9%).The cells were arrested in G 0/G 1 phase,but the numbers of S phase decreased obviously.CONCLUSION Tanshinone ⅡA can obviously inhibite growth of several tumor cell lines.NB4 cells is more sensitive to Tan ⅡA
