摘要
目的 研究骨髓增生异常综合征 (MDS)患者骨髓内T细胞亚群水平 ,T细胞产生IFN γ能力 ,及与造血干细胞凋亡的关系 ,探讨T细胞、IFN γ在MDS发病中作用。方法 骨髓单个核细胞经短期培养并激活T细胞 ,采用细胞内因子测定技术 ,用流式细胞术测定骨髓内T细胞亚群 ,T细胞IFN γ水平 ,RT PCR技术测定BMMNC凋亡基因Fasm RNA水平。结果 MDS患者骨髓内异常活化的T细胞 (CD3 + CD45RO+ 、CD4+ CD45RO+ 、CD8+ CD45RO+ 细胞 )增多 ,活化的T细胞产生IFN γ增多 ,并与Fas mRNA存在相关性。结论 MDS患者骨髓内活化的T细胞增多 ,这些细胞产生的IFN γ可能增加凋亡基因Fas的表达。
Objective To explore the relationship between IFN-γ among T lymphocyte subsets in Bone Marrow of myelodysplastic syndromes(MDS) and Fas gene and distribution of Tlymphocyte subsets,IFN-γ producing ability of T cell as well.Methods Following marrow cells culture,different T lymphocyte subsets in BM were categorized by monoclonal antibodies with 3 color fluorescence using flow cytometry,then the intracellular IFN-γ were determined.Fas mRNA of Bone Marrow mononuclear cells were examined by RT-PCR.Results The actived T lymphocyted(CD 3 +CD 45 RO +、CD 4 +CD 45 RO +,CD 8 +CD 45 RO +) in BM of MDS were higher than in controls;IFN-γ levels in BM of MDS increased;The IFN-γ produced by the actived T lymphocyted cells;Fas mRNA expression had relationship with the IFN-γ produced by the T cells.Conclusion The actived T lymphocyted in BM of MDS were higher,IFN-γ produced by the T cells The IFN-γ play an important role in hematopoitic cells apoptosis within patients of MDS.
