摘要
目的 :研究血管内皮生长因子 (VEGF)和明胶酶 -A(matrixmetalloproteinase -2 ,MMP -2 )的表达与口腔鳞癌转移的相关性及VEGF和MMP -2的关系。方法 :应用Boyden小室体外侵袭实验比较口腔鳞癌细胞系TSCCa和转移癌细胞系GNM的体外侵袭力 ;采用半定量逆转录聚合酶链反应 (RT -PCR)检测两细胞系中VEGF和MMP -2mRNA表达情况 ,同时用酶联免疫吸附实验 (ELISA)的方法定量测定口腔鳞癌TSCCa细胞系和颈淋巴转移癌GNM细胞系中VEGF和MMP -2的活性。结果 :Boyden小室体外侵袭实验表明转移癌细胞系GNM的体外侵袭力较强 ,其穿膜细胞相对百分率为 (40± 3.6 ) %,而口腔鳞癌细胞系TSCCa体外侵袭力相对较弱 ,其穿膜细胞相对百分率为 (16± 3.2 ) %,两者差异具有显著性 (P <0 .0 5 ) ;RT -PCR结果显示GNM细胞VEGF和MMP-2mRNA表达水平均较TSCCa细胞高 (P <0 .0 5 ) ;ELISA法显示颈淋巴转移癌GNM细胞系中VEGF和MMP-2的活性显著高于口腔鳞癌细胞系TSCCa(P <0 .0 5 )。结论 :口腔鳞癌细胞体外侵袭力与VEGF和MMP -2的表达正相关 ;在口腔鳞癌侵袭转移中 ,VEGF与MMP -2可能具有某种内在联系 ,有可能作为反映口腔鳞癌侵袭转移的生物学指标。
Objective:To investigate the in vitro invasion and the expression of vascular endothelial growth factor (VEGF) and matrix metalloproteinase-2(MMP-2) in oral squamous cell carcinoma cells,and to evaluate the correlation of expression of VEGF and MMP-2 in oral squamous cell carcinoma tumor metastasis. Method:Boyden chamber in vitro invasion assay was used to detected the invasive capacity in vitro in two oral squamous cell carcinomas cells TSCCa and GNM. The expression levels of VEGF and MMP-2 in two cell lines was evaluated by Semi-quantiative RT-PCR. The activities of VEGF and MMP-2 in two cell lines was detected by enzyme linked immunosorbent assay (ELISA). Result:Boyden chamber in vitro invasion assay indicated that the mean invasion percentage of GNM cells (40±3.6) was significantly higher than that of TSCCa cells (16±3.2)( P<0.05); Contrast to TSCCa cells, GNM cells expressed signficantly higher levels of mRNA of VEGF and MMP-2 by RT-PCR. ELISA showed that the activities of VEGF and MMP-2 in GNM cell lines was significantly higher than that of in TSCCa cell lines (P<0.05).Conclusion:The in vitro invasive ablity of oral squamous cell carcinomas cells appeared to be positive correlated to high expression to high expression of VEGF and MMP-2. There may be relationship between VEGF and MMP-2 in the process of metastasis of oral squamoua cell carcinoma. VEGF and MMP-2 may serve as a predictor of metastasis
