摘要
目的 揭示大脑中动脉闭塞 (MCAO)后丘脑钙调磷酸酶 (CaN)的时空变化规律 ,探讨CaN的作用机制。方法 制备大鼠大脑中动脉永久性闭塞模型 ,分别测定缺血后不同时间点病灶侧丘脑CaN的活性和含量。结果 缺血后 2 4h始丘脑CaN的含量下降且不恢复 ;CaN的活性在缺血后 2h和 4h减弱 ,6h始恢复至正常水平。可见 ,CaN的活性与含量分离。结论 大脑中动脉闭塞后丘脑CaN活性独特的时间变化规律显示其参与介导继发性丘脑损伤 ,可能具有毒性作用。
Objective To investigate the changes in the enzyme activity and q uantities of calcineurin(CaN) in the rat thalamus after middle cerebral artery occlusion,and to study its possible mechanism. Methods An immunoblotting technique and biochemical method was used to examine the activities and quantities of CaN following middle cerebral arter y occlusion (MCAO) in the ipsilateral thalamus of adult rats. Results Immediately after MCAO,CaN activity decreased temporar y at 2 hours and 4 hours,and then recovered;CaN quantity decreased at 24 hours after MCAO,and never recovered. Conclusion The unique time course of CaN activity may contribute to the mechanism of secondary thalamic degeneration after cerebral infarction in t he middle cerebral artery,and may play a toxic role in it.
