摘要
目的:探讨人慢性髓系白血病K-562来源的树突状细胞的免疫功能。方法:用细胞因子粒-巨噬细胞集落刺激因子(GM-CSF)及IL-4在体外诱导培养K-562细胞,获得树突状细胞(DC)检测其细胞表型,并观察其诱导的细胞毒T淋巴细胞(CTL)体外抗肿瘤效应。用ELISA法测定DC培养及DC与外周血单个核细胞共培养上清液中IL-12及IFN-γ的量。结果:联合GM-CSF及IL-4可诱导K-562细胞分化为树突状细胞,K-562 DC体外激活的CTL对K-562细胞具有特异性细胞毒活性;诱导DC培养上清及DC与外周血单个核细胞共培养上清测出一定量IL-12及IFN-γ。结论:人慢性髓系白血病K-562来源的树突状细胞表达抗原提呈细胞表型,具有诱导CTL反应和分泌IL-12以及促进T细胞分泌IFN-γ的作用。
Objective: To explore the immune function of K-562 dendritic cell (K-562 DC) derived from CML line. Methods: After being treated by GM-CSF and IL-4, K-562 cells were differentiated into K-562 DC. The phenotype of K-562 DC and the anti-tumor effects of CTL induced by K-562 DC were investigated. The level of IL-12 and IFN-'y in supernant were measured by ELISA. Results: The DCs were successfully differenfiated from K-562 cells. K-562 leukemic cells were effectively and specifically destroyed by CTLs induced by K-562 DC. Different levels of IL-12 were detected in the supernant of culturing DCs and IFN-r in the supernant of coculti-vation of DC and blood mononuclear cells. Conclusion: It was confirmed that DCs derived from chronic myeloid leukemic cells can express phenotype of antigen-presenting cells. They have the ability to induce CTLs to clear up leukemia cell and secrete IL-12 and increase IFN--ysecretion of T cells.
