恶性肿瘤组织肿瘤转移相关基因的表达与肿瘤细胞生物学特性的关系研究 被引量:1

摘要 目的:探讨各种肿瘤转移促进基因和肿瘤转移抑制基因编码蛋白在恶性肿瘤转移中的表达。方法:采用流式细胞术对56例肿瘤转移和61例肿瘤未转移患者瘤组织的CD44V5^+、CD44V6^+、cerbB-2^+、nm23^+、DCC^+和p16^+细胞检出率和DNA含量进行检测和分析。结果:恶性肿瘤患者瘤组织细胞的CD44V5^+、CD44V6^+和cerbB-2^+细胞表达率均显著高于正常对照组(P<0.01或P<0.05);而nm23^+,DCC^+,p16^+细胞表达率则显著低于正常对照组(P<0.01或P<0.05)。肿瘤转移者、多器官转移者、DNA异倍体者、SPF增高者、Apo降低者的CD44V5^+、CD44V6^+、cerbB-2^+细胞表达率均显著高于未转移者、单器官转移者、DNA二倍体者、SPF降低者、APO增高者(P<0.01或P<0.05);而nm23^+、DCC^+、p16^+细胞的表达率与此相反。结论:肿瘤转移相关基因编码蛋白在各种恶性肿瘤转移中均出现异常表达,但有一定的倾向性,且与瘤细胞的生物学特性(DNA倍性、细胞增殖活性、细胞凋亡水平)的关系均十分密切。 Objective: To study the expression of tumor metastasis enhancing genes and tumor metastasis suppressor genes coding proteins in tumor tissues from patients with malignant carcinoma. Methods: The expression rates of CD44V5 , CD44V6 , cerbB-2 , nm23, DCC, p16 cells and DNA contents in tumor tissues from 117 patients were measured and analyzed with malignant carcinomas by flow cytometry. Results: The cellular expression rates of CD44V5, CD44V6 and cerbB-2 in tumor tissues of patients with malignant carcinomas were higher than those of normal controls ( P < 0. 01 or P < 0. 05 ), while the cellular expression rates of nm23, DCC, p!6 were lower than the control (P < 0. 01 or P < 0. 05). The cellular expression rates of CD44V5 , CD44V6, cerbB-2 of patients with metastasis, multi-metastasis, DNA heteroploid, high SPF and low Apo were higher than those of patients with non-metastasis, single-metastasis, DNA diploid, non-high SPF , and non-low Apo( P < 0. 01 or P < 0. 05) . While the expression of nm23, DCC, p16 cells in tumor tissues from patients with malignant tumor were contrary to CD44V5, CD44V6, cerbB-2 Conclusion: The expression level of different tumor metastasis related genes coding protein in tumor tissues of patients with malignant tumor is obviously various but has certain tendency among different kinds tumors, which is closely related to the biological characteristics including DNA ploidy, proliferative activity and apoptosis level of tumor tissues.
