摘要 目的:观察碱性成纤维细胞生长因子(bFGF)对骨髓基质细胞的增殖及成骨活性的影响。方法:传代骨髓基质细胞的培养中加入不同浓度的bFGF,行细胞增殖和蛋白含量测定以及碱性磷酸酶的活性检测与钙结节的形成观察。结果:bFGF浓度在0~10ng/ml,蛋白含量及细胞数量随浓度的增高而增多,10ng/ml达到最大,浓度为100ng/ml反有下降。各组ALP活性没有降低,Von Kossa染色均为阳性。结论:bFGF可促进骨髓基质细胞的增殖,并保持骨髓基质细胞成骨的特性。 Objective: To observe the effect of bFGF on proliferation and ossify competence of BMSCs. Methods: The bFGF was added into the cultured medium of bone marrow stromal cell with different concentrations. Protein content and cell proliferation were measured in 3 days and 6 days respectively. Meanwhile ALP activity and the formation of mineralized nodules were detected. Results: Protein content and cell proliferation increased along with the increase of bFGF concentration among 0 ~ 10 ng/ml and at 10 ng/ml concentration of bFGF the cell proliferation arrived the submit, but at 100 ng/ml concentration of bFGF the cell proliferation decreased. The level of ALP didn' t play down and Von Kossa staining was positive in each group. Conclusion: bFGF can stimulate the proliferation of bone marrow stromal cells and preserve the ossify competence at the same time.
