摘要
合成了化学修饰剂 (对氨基苯甲酸对脒基苯酯盐酸盐 ) ,并通过透析法得到了 L ys-纤维蛋白溶酶原。提出了利用 L- L ysine Sepharose4 B柱以人纤维蛋白溶酶原、链激酶及化学修饰剂制备对氨基苯甲酰纤维蛋白溶酶原 -链激酶激活剂复合物 (APSAC)的新方法 ,本法的优点是 APSAC的收率及纯度均较高。
Amidinophenyl-4′-aminobenzoate hydrochloride was synthesized and used as the chemical modification agent for preparation of p-aminobenzoyl plasminogen streptokinase activator complex(APSAC). The Lys-plasminogen was obtained by dialysis of Glu-plasminogen against a lysine solution (10 mmol/L, pH 6.8). APSAC was prepared by chromatography on the L-lysine Sepharose 4B column loaded with Lys-plasminogen, streptokinase and the chemical modification agent. The advantage of this method was higher recovery and purity of the product.
