摘要 目的 :检测不同浓度胰岛素样生长因子Ⅱ (Insulin -likegrowthfactorⅡ ,IGF -Ⅱ )对电离辐射后成骨细胞增殖、功能和分化方面的影响 ,探讨不同剂量辐射后IGF -Ⅱ对成骨细胞的效应。方法 :成骨细胞通过酶消化法从SD大鼠颅骨中获取传代 ;将第 3代细胞分别进行 0、10 0、4 0 0、6 0 0和 90 0cGy的γ线辐射 ,辐射后的细胞在含有不同浓度IGF -Ⅱ的培养液中培养 6d ,观察及检测成骨细胞的增殖、功能及分化等情况。结果 :经电离辐射后 ,细胞的增殖、碱性磷酸酶活性及骨钙素含量均受到抑制。IGF -Ⅱ可有效降低 10 0cGy辐射对增殖的抑制效应 ,1,10 ,和 10 0 μg/L的IGF -Ⅱ都改变了 4 0 0cGy时的辐射效应 ,其中以 10 μg/L的浓度时最明显。 结论 :IGF -Ⅱ能有效促进成骨细胞辐射损伤的恢复 ,而这种作用有浓度依赖性 ,并取决于辐射损伤的程度。 Objective:To investigate the effects of Insulin-like gr ow th factor (IGF) -Ⅱon the proliferation, function and differentiation in the rat osteoblast-like cells(ROB) after ionizing radiation in vitro.Methods :The kind of osteoblast-like cells was isolated from the calvariae of neonatal (one-day-old) SD rats by sequential enzymatic digestion.The third pass age of the cells was used for all the experiments described below. All the sampl es were exposed to the specified γ-ray dose (100, 400, 600, or 900cGy) on the t hird day in culture after being seeded. The culture medium was replaced after ra diation by RPMI1640 further supplemented with IGF-Ⅱ.MTT,ALP activity and BGP wa s determined.Results:Cell growth decreased with increasing dose of radiation, so did the ALP activity and BGP. IGF-Ⅱat any concentration enhanced the growth of the cells after 100cGy radiation.1,10, and 100μg/L IGF- Ⅱ induced significant changes in the values compared with those in the correspo nding control cultures with the radiation dose of 400cGy.10μg/L is believed to be much more significant.Conclusions:Incubation with IGF- Ⅱcan increase radioresistance or repair of radiation-induced cells damages.The effects of IGF-Ⅱon the recovery of ROB after radiation are dose-dependent, and of 4 different concentration, 10μg/L is the strongest.
