摘要
目的 :观察气虚证模型大鼠鼻咽组织B区基因表达谱特征及其对治疗的反应。方法 :药物加疲劳法在Wistar大鼠制作气虚证模型 ,随机分为治疗组与对照组 ,以AtlasTMcDNA阵列法检测各组动物鼻咽组织B区基因表达谱 ,比较分析其表达谱差异。结果 :模型组动物表现I -kBα链、MAPK1、MAPK2、GNB2基因表达上调 ,MEK5、PLCδ、RGP(Rab相关GTP结合蛋白 )基因表达下调 ;治疗 1周后 ,表达上调的MAPK1、MAPK2二基因活性恢复正常 ,其余 5个基因表达活性仍维持治疗前活性状态。结论 :单纯气虚证动物出现调制子、效应子和细胞内转导子基因表达活性异常 ,细胞内信号传导通路发生障碍。经针对性治疗后 ,这些障碍可以逐渐恢复正常。
Objective:To investigate the features of gene expression profilingon B area of Atlas TM cDNA array and their respones to treatment in treatment in nasopharyngeal tissues among rats with Qi deficiency.Methods:Qi deficiency was induced among rats with the method of herbal decoction perfused gastrically and supplemented by forced swimming in deep pool.The expressive features of genes on B area were determined in nasopharyngeal tissues of tested rats on Atlas TM cDNA expression array.Results:The genes showing up regulated expressive activities included I-κ B α chain,MAPK1,MAPK2 and GNB2,while those showing down regulated expressive activities were MEK5,PLCδ1 and Rab related GTP-binding protein(RGP)among the rats of modeling group.In contrast,the animals in treating group showed normally recovered expressive activities of MAPK1 and MAPK2,while the remainders were still remained in an abnormal status of expressive activities as in modeling group.Conclusions:Rats with Qi deficiency may show abnormal gene expressive activities of modulators,effectors and transducers in the intracellular signal transmitting pathway.In contrast,these abnormally expressed gene activities will be brought into a normal status gradually after effective treatment.
