摘要
目的:检测骨关节炎(OA)软骨组织中BAX基因的表达及其与骨关节炎临床分期的关系。方法:应用逆转录PCR(RT-PCR)方法检测6例正常软骨组织、15例中期骨关节炎患者和21例晚期骨关节炎患者软骨组织BaxmRNA的含量;应用定量图像分析比较相对含量。结果:Bax基因在中期骨关节炎患者的软骨组织中表达阳性率为60%(9/15);明显低于晚期骨关节炎患者91%(20/22),P<0.01。且基因表达量与骨关节炎病变程度存在程度一致性;结论:Bax mRNA的表达与骨关节炎的病变程度相关,并通过对细胞凋亡的调控参与骨关节炎的发生、发展。
Objective: To investigate the expression and significance of apoptosis-relating Bax mRNA in osteoarthritis and the relations between their expression and clinicoplathologic characteristic. Methods: Bax mRNA was analyzed in 42 OA patients, including 6 normal cartilage, 15 intermediate stage OA and 21 later stage OA tissues,by reverse transcriptase polymerase chain reaction(RT-PCR). Results: There was significant difference of expression of Bax mRNA between early OA(11/15) and terminal OA( 18/22). The positive rates of Bax mRNA expression in early OA patients, terminal OA, normal cartilage tissures was 73. 5% (11/15) ,81.8% (18/22) and 33.3% (2/6) respectively. Their dufference was sugnificant(P<0.01). And the level of Bax mRNA expression had significant coherence with the degree of OA. Conclusion: The expression level of Bax may play a crucial role in the OA development, indicating that controlling Bax may provide a target for the gene therapy for OA.
