TFP促进粟酒裂殖酵母(Schizosaccharomyces pombe)细胞增殖机理的探讨

摘要 采用激光扫描共聚焦显微镜观察的方法,以Fluc-3负载Schizosaccharomyces Pombe细胞,荧光强度反映胞质游离Ca2+浓度,结果表明,在无钙培养基中生长的S.Pombe胞内游离Ca2+浓度低于在含10μmol/L外钙培养基中的S.pombe细胞,而一定浓度三氟拉嗪(TFP)处理过的S.pombe胞内游离钙浓度则有明显增加;用与TFP在对酵母质膜ATPase活性有拮抗效应的无机硫酸盐处理S.pombe细胞,发现可抑制其增殖,通过原子吸收光谱法测得胞内游离钙含量降低,因此认为TFP通过作用于质膜、促进Ca2+内流刺激裂殖酵母细胞的增殖。 Schizosaccharamyces pombe cells loaded with Fluo-3 were observed under laser scanning confocal microscope (LSCM). The result showed that the cytosolic free Ca2+ concentration in S. pombe cells cultured in Ca2+ -free medium was lower than cells cultured in medium containing 10μmol/L Ca2+ , while Ca2+ concentration in cells treated with 50μmnol/LTFP increased apparently. We also found that inorganic sulfate which arrested the growth of S. pombe decreased the cytosolic free Ca2+ concentration. Therefore, we conclude that TFP promotes the proliferation of S. pombe cell by stimulating Ca2+ influx.
