摘要
目的:探讨c-fos在阿尔茨海默病(Alzheimer'sdisease,AD)模型大鼠和血管性痴呆(vasculardementia,VD)模型大鼠脑中表达的特征,并分析是否存在差异及其意义。方法:实验使用SD雄性大鼠,体质量350~520g;先对所有大鼠进行迷宫训练,筛选出达到学会标准的大鼠,再将其随机分成对照组(3只),AD模型组和VD模型组。手术切断大鼠左侧穹窿海马伞(FF)建立AD大鼠模型,以在颅骨外固定小磁铁吸附经舌下静脉注入Fe3O4粉进而堵塞脑血管复制缺血性VD大鼠模型。在经迷宫检查证实上述模型建成后的不同时段,对含有海马结构的脑组织切片进行Fos蛋白的免疫组化染色。结果:Fos样免疫反应活性(Fos-LI)在AD大鼠的海马及大脑皮质均有增强,并随建模后时间的延长呈逐渐增强的趋势,在VD大鼠脑中Fos-LI则以皮质增强为主且只在建模初期有增强,1周时即已恢复正常。结论:c-fos在AD及VD模型大鼠脑中的表达存在空间及时程上的差异,反映了c-fos功能的多样性及复杂性。
AIM:To investigate the characteristics of c-fos expression in the brain of ra t Alzheimer' s disease(AD)and vascular dementia(VD)models.METHODS:Twenty-one male SD rats which had passed Y-maze test were divided randomly into control group(n=3),AD model group(n=9)and VD model group(n=9).AD models were established by compl ete severing of the unilateral fimbria hippocamp i of the rats,and VD model by Fe 3 O 4 powder injection into the sublingual vein and fixation of a magnet on the s kull surface of the rat.At different time points after model establishment as veri-fied by Y-maze test,Fos immunohisto chemical staining of the slices of th e brain tissue including the hippocam pus was performed.RESULTS:The Fos-like immunoactivity(Fos-LI )increased markedly in the cortex and hippocampus of rat AD models,which intensified gradually wit h the elapse of time.Fos-LI also incre ased on the second day after VD model was induced,on the day 7,normal condition was resumed,and the Fos-LI was far more intense in the cortex than in the hippocampus.
