脑梗死患者脑小动脉平滑肌细胞表型转化与血管修复的相关性 被引量:9

摘要 目的:观察两种血管收缩功能相关蛋白在脑梗死患者小动脉血管平滑肌细胞(vascularsmoothmusclecell,VSMC)中免疫组织化学表达变化,以期了解脑小动脉病变中VSMC的表型转化及细丝蛋白在表型转化中的意义,指导临床小动脉硬化型脑卒中的准确防治。方法:选取尸解脑标本,分为脑梗死组及对照组;免疫组化显示脑小动脉α平滑肌肌动蛋白及细丝蛋白的表达变化,并进行定量分析。结果:对照组α平滑肌肌动蛋白及细丝蛋白表达强度(PU)分别为53.50±2.00和34.21±2.36;梗死组为57.40±2.43和36.10±1.51,与对照组相比F值为15.273和4.592,P值为0.001和0.046,梗死组阳性表达强度增高,差异有统计学意义。讨论:硬化小动脉中膜数目减少。内膜下VSMC显著增生,并可能同时存在合成及收缩表型VSMC,分别代偿性的高表达细丝蛋白及α平滑肌肌动蛋白,以维持血管构型及收缩能力。 AIM:To observe the expressions of two rel ated vasoconstriction proteins in the arteriolar smooth muscle cells(VSMC)in patients with cerebral infarc-tion by using immunohistochemical m ethod so as to understand the phenoty pe switching in arteriolar VSMC and the meaning of filamin in the phenotype switching for exactly preventing arteriosclerotic stroke in clinic.METHODS :Brain tissue samples were removed fr om corpses with cerebral infarction and randomized into cont rol group and cerebral infarction group.By using immunohistochemical method,the expression of α-SM-actin and filamin in cerebral arteriolar VSMC were quantitatively measured and analyzed.RESULTS:The PU values of positiveα-SM-Actin and filamin were 53.50±2.00and 34.21±2.36for the control g roup,and 57.40±2.43and36.10±1.51for the cerebral infarction group(F=15.273,4.592,P=0.001,0.046).
