金铁锁总甙对小鼠细胞免疫功能的影响 被引量:22

Immunomodulatory effects of total glycosides of Psammosilene tunicoides on cell immunity in mice

摘要 目的 阐明金铁锁总甙 (TGP)对小鼠细胞免疫功能的影响。方法 以 80和 2 5 0mg/kg环磷酰胺分别诱导小鼠迟发性超敏反应 (DTH)增强和抑制模型 ,观察不同剂量的TGP对小鼠DTH的影响。以 0 6 %巯基乙醇酸钠诱导、制备和培养小鼠腹腔单层巨噬细胞 ,观察不同浓度的TGP对巨噬细胞产生IL - 1的影响。以ConA诱导小鼠脾淋巴细胞和胸腺淋巴细胞增殖反应 ,观察不同剂量TGP在给药 (ig)的第 5、10、15天对小鼠脾淋巴细胞的增殖反应和生成IL - 2的影响。结果  6 0~ 10 0mg/(kg·d)的TGP均能显著提高细胞免疫抑制小鼠的DTH反应 (P <0 0 1)。 2 0~ 10 0mg/(kg·d)能显著下调细胞免疫增强小鼠的DTH(P <0 0 1)。 3μg/ml的TGP能显著提升小鼠巨噬细胞产生IL - 1的水平。 2 0~ 80mg/(kg·d)的TGP对小鼠脾淋巴细胞增殖反应均有明显的促进作用 ,80mg/(kg·d)的促进作用最为显著 (P <0 0 1) ,10 0mg/(kg·d)时 ,这种促进作用减弱。 2 0~ 80mg/(kg·d)的TGP在给药的第 15天均能显著提升小鼠脾淋巴细胞产生IL - 2的水平 ,以 6 0mg/(kg·d)最为显著 ,80mg/(kg·d)以上 ,这种提升作用减弱。结论 适当剂量的TGP既是小鼠细胞免疫的增强剂 。 Objective To elucidate the immunomodulatory effects of total glycosides of Psammosilene tunicoides (TGP) on the cell immunity in mice.Methods Enhanced and suppressed delayed-type hypersensitivity (DTH) in mice was induced with cyclophosphamide at doses of 80 and 250 mg/kg,respectively to determine the effects of different dosage of TGP on DTH.The monolayer celiac macrophages of mice induced with 0.6% sodium thioglycolic acid and were cultured to observe the impact of macrophages by different concentrations of TGP on interleukin-1 secretion.Spleen and thymus lymphocyte proliferation was induced by concanavalin A to examine the influence of different doses of TGP at different days of administration on the lymphocyte proliferation and secretion of interleukin-2.Results 60~100 mg/(kg·d) of TGP significantly enhanced the DTH in immuno-suppressed mice (P<0.01),and 20~100 mg/(kg·d) of TGP sigmificantly decreased the DTH in immuno-increased mice (P<0 01).3 μg/ml of TGP significantly increased the secretion of IL-1 by macrophages.20~80 mg/(kg·d) of TGP significantly facilitated the lymphocyte proliferation in mice,with the optimum dose at 80 mg/(kg·d) (P<0 01).This facilitation,however,was weakened at 100 mg/(kg·d).20~80 mg/(kg·d) of TGP significantly increased the secretion of IL-2 by spleen lymphocytes after 15 days of administration, with the optimum dose at 60 mg/(kg·d),but the secretion of IL-2 tended to decline at the dose over 80 mg/(kg·d).Conclusions Adequate dose of TGP not only enhances but also modulates the cell immunity of mice.
