骨巨细胞瘤VEGF和P53蛋白表达及其与微血管密度和肿瘤复发转移的关系

Association of VEGF and P53 protein expression with microvessel density and recurrence and metastasis of giant cell tumor of bone

目的 探讨骨巨细胞瘤中血管内皮生长因子 (VEGF)和P5 3蛋白表达与微血管密度 (MVD)、肿瘤分级和复发转移的关系。方法 应用LDP法免疫组织化学技术。对 5 1例骨巨细胞瘤进行VEGF、P5 3蛋白和FⅧRAg染色 ,检测VEGF和P5 3蛋白的表达 ,FⅧRAg显示肿瘤组织中的MVD ,并进行相对定量研究。结果 骨巨细胞瘤中VEGF高表达率为 49 0 2 % (2 5 /5 1)。VEGF高表达率在Enneking 1级与 2级或 3级之间差异有显著性 (P <0 0 5 ) ,在复发组与无复发组和转移组与无转移组之间差异也都有显著性 (P <0 0 5 ) ,与病理分级无关。骨GCT中 12例P5 3蛋白有阳性表达 ,阳性率为2 3 5 3 %。P5 3阳性率在Enneking 1级与 2级或 3级之间差异有显著性 (P <0 0 5 ) ,在复发组 (3 3 3 3 % )与无复发组(9 5 2 % )和转移组 (5 0 0 % )与无转移组 (2 0 % )之间差异都有显著性 (P <0 0 1)。在骨巨细胞瘤复发组和转移组的MVD都分别高于未复发组或无转移组 ,两组间差异都有显著性 (P <0 0 5 )。VEGF与P5 3呈显著正相关 (r =0 697) ;VEGF和P5 3与MVD表达也呈显著正相关 (r=0 792 ,r =0 787)。结论 VEGF、P5 3在骨巨细胞瘤中的表达与Enneking分级有一定关系 ,与微血管密度相一致 ,并与骨巨细胞瘤复发转移有关 ,VEGF、P5

Objective To Study the association of vascular endothelial growth factor(VEGF) and P53 protein expression with microvessel density(MVD) and recurrence and metastasis in giant cell tumor(GCT) of bone Methods VEGF expression and P53 protein and F Ⅷ RAg in 51 cases of GCT of bone were studied by immunohistochemistry staining,and the relative level analysis was used to determine deal the positive expression and MVD Results In 51 cases the over expression rates of VEGF and P53 protein in GCT of bone were 49 02%(25/51),23 53%(12/51)respectively Over expression of VEGF and P53 protein which had no relation with Jaffe's pathological grade in GCT of bone was found to be related to Enneking classification to some extent Over expression rate of VEGF and P53 protein of Enneking grade 1 of GCT was significantly lower than those of grade 2 or grade 3( P <0 05) Over expression rates of VEGF and P53 protein in recurrence and metastasis were significantly higher than those of the control group( P <0 05) There was a significant positive correlation between over expression rates of VEGF and P53 protein and microvessel density in GCT of bone Conclusion Over expression of VEGF and P53 protein in GCT of bone is related with recurrence and metastasis in GCT of bone along with its MVD Therefore,VEGF and P53 protein and MVD may be important reference parameters in predicting the prognosis of GCT of bone
