摘要
目的 研究不同剂量顺铂 (Cisplatin ,CP)诱导大鼠肾损伤的组织病理学变化 ,建立急性肾功衰竭动物模型。方法 Wistar成年大鼠 ,腹腔注射CP ,模型 1组 5mg/kg ,模型 2组 7mg/kg ,给药后于不同时间取肾 ,常规石蜡切片 ,HE染色和PAS反应。结果 两组鼠均引起肾皮质损伤 ,可见肾小球肿大 ,近端小管上皮浊肿、空泡形成乃至细胞脱落死亡及毛细血管基膜损伤 ,以 7mg/kg为甚。结论 CP 7mg/kg能复制出典型的急性肾性肾功衰竭组织病理学改变 。
Objective: To study the histopathologic changes on kidney damage of rats caused by various volume of dosage cislplatin (CP) of rats, and establish acute renal failure animal model Methods: CP was injected into the intraperitoneal of adult Wistar rats with dose 5 mg/kg in CP1 group,7 mg/kg in CP 2 group For observation with microscope, the kidneys were taken out at every time and undergone route paraffin sections, HE stained and periodic acid Schiff reaction (PAS reaction) Results: Cortex of the kidneys was damaged The renal corpuscle swelled, vacuoles even necrotic cells of proximal tubules were observed throughout the cortex, basement membrane of the capillary of the renal corpuscle and proximal tubules were also damaged, the rats in CP 2 group weremore serious Conclusion: The results show that the typical histopathology changes of acute renal failure induced by CP are duplicated with dose 7 mg/kg, so it can be a successful animal model for relavant studies
